1. Define the Target Product Profile
Before equipment settings are selected, the development team defines intended use, target market, active amount per strip, film dimensions, disintegration expectation, flavor, appearance, packaging and shelf-life goal. These requirements determine whether the concept is technically realistic.
2. Prepare the Film-Forming System
Polymers, plasticizers, sweeteners, flavors and functional ingredients are dispersed or dissolved in a qualified liquid system. Order of addition matters: poor wetting can create agglomerates, while excessive aeration can produce bubbles and weak points. Viscosity and solids content must remain suitable for uniform casting.
3. Deaerate and Condition
Entrained air is reduced before casting. The solution may also need controlled hold time so hydration and viscosity stabilize. Long uncontrolled holds are avoided because flavors can volatilize and suspended particles can settle.
4. Cast a Uniform Wet Film
A metering system places the film-forming mass onto a carrier at a controlled wet thickness. Web speed, gap, viscosity and temperature interact. In-process checks look for streaking, edge effects, pinholes and coating-weight drift.
5. Dry Within a Controlled Window
Drying removes solvent while preserving film integrity. Too aggressive a profile can cause curling, bubbles or surface skinning; too gentle a profile can leave excess residual moisture and poor handling. Heat-sensitive ingredients and volatile flavors may constrain the acceptable window.
6. Slit, Cut and Verify Unit Dose
The dried web is converted into individual films. Dimensions and weight are controlled because they are linked to delivered amount when the active is uniformly distributed. Finished testing may include identity, assay, unit uniformity, disintegration, moisture, mechanical properties and microbiological quality as appropriate.
7. Seal in Barrier Packaging
Individual sachets help protect hygroscopic films from humidity, oxygen and handling. Material selection and seal integrity are part of shelf-life design, not an afterthought.
Scale-Up Risk Controls
- Establish acceptable viscosity and solids ranges rather than a single lab value.
- Link coating weight and film area to dose uniformity.
- Map drying conditions against residual moisture and physical properties.
- Qualify foil structure and seals under stability conditions.
- Use change control when raw-material grade or supplier changes.
Is solvent casting the only ODF process?
It is common for many oral films, but other technologies may be used. Process selection depends on the ingredients, facility and product classification.
Why is unit-dose packaging common?
Thin films are sensitive to moisture and handling. Individual barrier packs protect performance and provide convenient dosing.
Can any tablet formula be converted into a strip?
No. Loading, solubility, taste, stability and film mechanics can make a tablet formula unsuitable without significant reformulation.
